Mechanism of action (MOA)

1. Pregabalin: Binds to α2–δ subunits of voltage gated ion channels in CNS, thereby reducing neuronal excitability
2. Duloxetine: A serotonin-norepinephrine reuptake inhibitor, thereby enhancing descending pain inhibition2. Offers faster central pain relief when compare with Nortriptyline
